Background ErbB2, also called Neu or Her2, is a transmembrane glycoprotein encoded by human Her-2 gene, which is closely related to the epidermal growth factor receptor protein. ErbB2 is a 185 kDa glycoprotein that contains an extracellular ligand-binding domain and intracellular tyrosine kinase activity. ErbB2 is widely expressed in low levels in epithelial cells of normal human tissues and over-expressed in some type of cancers. ErbB2 heterodimerizes with ErbB1/ErbB2, ErbB3, or ErbB4 to form higher affinity signaling complexes. The ErbB2/ErbB3 heterodimer is expressed in the majority of breast, skin, ovary and gastrointestinal tumors and transduces a highly mitogenic signal in response to neuregulin 1 (NRG1) or NRG2. In these malignancies the HER-2 oncoprotein overexpression is correlated with a poor prognosis. Soluble forms of ErbB2 can be generated by proteolytic shedding or by alternative splicing. A 110 kDa soluble fragment of ErbB2 can be released by protease ADAM10 from the cell surface. Serum levels of soluble ErbB2 are elevated in individuals with Her2+ metastatic breast tumors and may be associated with survival.
