Myogenin is a member of the MyoD family of myogenic basic helix-loop-helix (bHLH) transcription factors that also includes MyoD, Myf-5, and MRF4 (also known as herculinor Myf-6). MyoD family members are expressed exclusively in skeletal muscle and play a key role in activating myogenesis by binding to enhancer sequences of muscle-specific genes. The regulatory domain of MyoD is approximately 70 amino acids in length and includes both a basic DNA binding motif and a bHLH dimerization motif. MyoD family members share about 80% amino acid homology in their bHLH motifs. Anti-myogenin labels the nuclei of myoblasts in developing muscle tissue, and is expressed in tumor cell nuclei of rhabdomyosarcoma and some leiomyosarcomas. Positive nuclear staining may occur in Wilms’ tumor.||Applications: |Suitable for use in Western Blot, Immunoprecipitation, Immunohistochemistry. Other applications not tested.||Recommended Dilution:|Western Blot: 0.5-1.0ug/ml|Immunoprecipitation: 0.5-1ug/500ug protein lysate|Immunohistochemistry: Frozen & Formalin-fixed: 0.5-1.0ug/ml for 30 minutes at RT|(Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 minutes|Optimal dilutions to be determined by the researcher.||Positive Control:|Rh-30 cells. Skeletal muscle or rhabdomyosarcoma.||Storage and Stability:|Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. |For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.||Note: Applications are based on unconjugated antibody.
