Protocol Summary:

  • Transfer standards or diluted samples into the transparent microtiter plate provided.
  • Add the DetectX® Creatinine Reagent to each well, ensuring thorough mixing.
  • Incubate the mixture at room temperature for 30 minutes to allow the color-generating reaction between the reagent and creatinine.
  • After incubation, use a plate reader to detect the signal at 490nm.
  • Calculate creatinine concentration using the standard curve and intensity measured. The assay uses a kinetic absorbance method to mitigate interference from colored compounds in the sample.

Background:

Creatinine is a breakdown product of phosphocreatine (p-creatine), a molecule important for energy storage in muscles. During muscle contractions, phosphocreatine breaks down, releasing energy and forming creatinine. This conversion process, which happens non-enzymatically, allows creatinine to diffuse into the bloodstream and be excreted by the kidneys. The rate of conversion can be influenced by factors like temperature and pH.

Measuring serum creatinine is a standard method for assessing renal function. Elevated levels of creatinine in the blood usually indicate significant kidney damage. The creatinine clearance test, which takes into account factors such as age, sex, weight, and race, is a reliable indicator of kidney function. Additionally, creatinine levels can be impacted by factors such as muscle mass, diet (especially meat consumption), certain drugs, and dehydration. Therefore, understanding creatinine levels in the context of these factors is essential in medical diagnostics.
