| Background | LYVE1 (lymphatic vessel endothelial hyaluronan receptor-1) is the lymphatic vessel endothelial hyaluronic acid receptor located in lymph nodes and in the luminal/abluminal surfaces of lymphatic vessels. It is one of the most specific and widely used mammalian lymphatic endothelial markers. LYVE-1 exists as disulfide-linked homodimer, can interact with PDGFB/IGFBP3 and also forms transient ternary complex with PDGFB and PDGFRB in trans-Golgi network (TGN). LYVE1 is a single-pass type I membrane protein localized in plasma membrane and in vesicles near extranuclear membranes which represents TGN and endosomes or pre-lysosomeal compartments. LYVE1 undergoes ligand-dependent internalization and recycling at the cell surface, and executes ligand-specific transporter trafficking between intracellular organelles/TGN and plasma membrane. LYVE1 plays a key role in autocrine regulation of cell growth mediated by growth regulators containing cell surface retention sequence binding. LYVE1 behaves as hyaluronan transporter, either mediating its uptake for catabolism within lymphatic endothelial cells themselves, or its transport into the lumen of afferent lymphatic vessels for subsequent re-uptake and degradation in lymph nodes. LYVE-1 is very efficient in the passage of lymphocytes as well as the tumor cells into the lymphatics. |
