This ELISA kit uses the Competitive-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with Universal 8-OHdG. During the reaction, Universal 8-OHdG in the sample or standard competes with a fixed amount of Universal 8-OHdG on the solid phase supporter for sites on the Biotinylated Detection Ab specific to Universal 8-OHdG. Excess conjugate and unbound sample or standard are washed away, and Avidin-Horseradish Peroxidase (HRP) conjugate are added to each micro plate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns from blue to yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of Universal 8-OHdG in tested samples can be calculated by comparing the OD of the samples to the standard curve.
8-OHdG (8-hydroxy-deoxyguanosine, also known as 8-OHDG) is an oxidizing adduct produced by reactive oxygen species such as hydroxyl radicals, singlet oxygen, etc. attacking the 8th carbon atom of guanine base in DNA molecules. It is a key biomarker of oxidative DNA damage, which can reflect the degree of oxidative damage and repair, as well as the relationship between oxidative stress and DNA damage. The detection of 8-OHdG is of great significance for the study of degenerative diseases, aging mechanism, carcinogenesis mechanism, and the relationship between environmental toxicants and oxidative stress. It can also be used to evaluate the effectiveness of antioxidants in treating oxidative DNA damage. At present, 8-OHdG has been widely used in experimental research and clinical diagnosis, especially in the clinical diagnosis and treatment of cardiovascular diseases, diabetes, neurodegenerative diseases (such as Parkinson's disease, Alzheimer's disease) and cancer.
