-20°CGab2 (Grb2-associated binder) is the latest member of a new family of multi-functional adapter molecules containing a Pleckstrin Homology domain, defined initially by the cloning of Gab1. Gab2 is broadly expressed as a 90-100kD protein. It contains two putative SH3 binding motifs and is tyrosine phosphorylated in response to a range of cytokine, growth factor, and antigen stimulation. Gab2 associates with SHP2, the p85 subunit of PI3K, Grb2, and SHC demonstrating its involvement in multiple signaling pathways. Gab 2 transfection experiments show that Gab2, like Gab1, stimulates Erk kinase activity following EGF-stimulation or co-transfection with activated Ras. By contrast, while Gab1 increases activated Ras or EGF-mediated transactivation of Ekl-1 responsive genes, Gab2 has an inhibitory effect. Thus, Gab2 may function as a negative regulator of cytoplasmic signaling to the nucleus. ||Applications:|Suitable for use in Western Blot. Not recommended for Immunoprecipitation. Other applications not tested. ||Recommended Dilution:|Western Blot: 0.5-2mg/ml. Detects GRB2 (23–25kD) in RIPA lysates of non-stimulated human A431 carcinoma cells. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. ||Note: Applications are based on unconjugated antibody.
