Adenosine Deaminase (ADA) Assay Kit for detection the activity of ADA in serum, plasma or tissue homogenates using a microplate reader. Adenosine deaminase (ADA) can hydrolyze the substrate adenosine to form hypoxanthine riboside, which is hydrolyzed by purine riboside phosphatase to produce hypoxanthine and phosphate ribose. Under the action of xanthine oxidase, hypoxanthine produces hydrogen peroxide, which produces a red colored reaction product under the action of peroxidase, 4-aminotepyrine and color source. The reaction product has a maximum absorption peak at 550 nm and the change in absorbance over time is proportional to the activity of ADA.
